Overexpression of RhoB in MCF-7 cells led to enhanced binding of histone deacetylase 1 (HDAC1) on the MMP-9 promoter. We found that the putative RhoB binding site in the promoter region of MMP-9 gene was indispensable for RhoB-mediated transcriptional regulation of MMP-9. In addition, overexpression of RhoB in MCF-7 cells promoted cell motility and invasiveness. The inhibition of cell growth could be attributed to increased caspase-3 activation.

Results showed that RhoB was upregulated in human breast cancer cell lines and tissues and overexpression of RhoB in MCF-7 cells inhibited cell proliferation and induced cell apoptosis.
